Phenotypic profle of monocyte-macrophage lineage cells as a function of respiratory epithelium status

چکیده

The mechanism of the relationship between pretumor changes in bronchial respiratory epithelium and risk progression non-small cell lung cancer (NSCLC) remains unclear. It has been suggested that reactive mucosa NSCLC may be caused by functional status monocytic-macrophage cells as important participants infammation, which determines both premalignant malignant progression. purpose study was to investigate phenotypic profle peripheral blood monocytes macrophages induced from vitro depending on state patients. Material Methods . included 39 patients with newly diagnosed NSCLC. Based morphological examination small bronchi taken at distance 3–5 cm tumor, were divided into following groups type changes: no (n=6), isolated basal hyperplasia (BCH) (n=13), combination BCH squamous metaplasia (SM) (n=3), unchanged focal (n=17). features -induced assessed before treatment using fow cytometry. Results prior start anticancer associated monocytes, but not them. Distortion response polarizing stimuli observed patients: cultured responded M1 M2 inducers (LPS IL-4, respectively) a phenotype shift M2, while CD206 marker expression varied presence changes. Conclusion anti-tumor treatment, macrophages.

Generation of Mature Monocyte-Derived Dendritic Cells in the Presence of Heparin and Monocyte Conditioned Medium: Phenotypic and Functional Comparison

Background: Dendritic cells (DC) induce tumor or pathogen-specific T cell responses in humans. Several laboratories have developed culture systems, including maturation factors for human DC from peripheral blood monocytes. We comprehensively compared standard maturation stimulus, an autologous monocyte-conditioned medium (MCM), with heparin for their ability to promote uniformly mature DC that ...

Regulation and function of the E-cadherin/catenin complex in cells of the monocyte-macrophage lineage and DCs.

E-cadherin is best characterized as adherens junction protein, which through homotypic interactions contributes to the maintenance of the epithelial barrier function. In epithelial cells, the cytoplasmic tail of E-cadherin forms a dynamic complex with catenins and regulates several intracellular signal transduction pathways, including Wnt/β-catenin, PI3K/Akt, Rho GTPase, and NF-κB signaling. Re...

Targeting the Monocyte–Macrophage Lineage in Solid Organ Transplantation

There is an unmet clinical need for immunotherapeutic strategies that specifically target the active immune cells participating in the process of rejection after solid organ transplantation. The monocyte-macrophage cell lineage is increasingly recognized as a major player in acute and chronic allograft immunopathology. The dominant presence of cells of this lineage in rejecting allograft tissue...
